Here is a step-by-step guide on how to calibrate a pipette.

  • Collect the necessary tools, such as the pipette, tips, beaker, thermometer, weigh boats, and distilled water.
  • Measure the water temperature. You will use the measurements during calculations to check calibration.
  • Use a scale with microgram readability to weigh the weigh boat. Ensure to set it to zero after closing the balance door.
  • Aspirate and dispense the set volume three times to pre-rinse the pipette tip. Ensure to remove any liquid.
  • Next, ensure to aspirate the calibration volume without bubble formation and dispense the liquid into the weight boat. Record the reading on the scale and repeat this ten times.
  • Use the equation V = W x Z to calculate the dispensed volume. W (water weight), Z (Z-factor), and V = dispensed water calculated volume.
  • Calculate the mean of the ten trials.
  • Use the equation A = 100 x Vavg/V0. Vavg (average calculated volume), V0 (volume you were dispensing), and A (pipette accuracy).
  • The pipette is calibrated if the range is between 99 and 101%.
